Действие комедии происходит в сказочной для англичан шекспировского времени стране — Иллирии.
Герцог Иллирии Орсино влюблен в юную графиню Оливию, но она в трауре после смерти брата и даже не принимает посланцев герцога. Равнодушие Оливии только разжигает страсть герцога. Орсино принимает на службу молодого человека по имени Цезарио, красоту, преданность и тонкость чувств которого успевает оценить всего за несколько дней. Его-то он и посылает к Оливии рассказать о своей любви. В действительности Цезарио — девушка по имени Виола. Она плыла на корабле вместе с любимым братом-близнецом Себастьяном и после кораблекрушения случайно оказалась в Иллирии. Виола надеется, что ее брат тоже спасся. Девушка переодевается в мужскую одежду и поступает на службу к герцогу, в которого сразу же влюбляется. За спиной герцога она говорит: «Мне нелегко жену тебе добыть; / Ведь я сама хотела б ею быть!»
Затянувшийся траур Оливии совсем не нравится ее дяде — сэру Тоби Белчу, весельчаку и гуляке. Камеристка Оливии Мария передает сэру Тоби, что ее госпожа очень недовольна кутежами и попойками дяди, а также его собутыльником сэром Эндрю Эгьючиком — богатым и глупым рыцарем, которому сэр Тоби морочит голову, обещая выдать за него племянницу, а тем временем беспардонно пользуясь его кошельком. Сэр Эндрю, обиженный пренебрежением Оливии, хочет уехать, но сэр Тоби, льстец и балагур, уговаривает его остаться еще на месяц.
Когда у дома графини появляется Виола, ее с большим трудом пропускают к Оливии. Несмотря на красноречие и остроумие, ей не удается добиться успеха своей миссии — Оливия отдает должное достоинствам герцога (он «несомненно, молод, благороден, / богат, любим народом, щедр, учен» ), но не люби! его. Зато юный посланец достигает совершенно неожиданного для себя результата — графиня очарована им и придумывает уловку, чтобы заставить его принять в дар от нее перстень.
Брат Виолы Себастьян появляется в Иллирии в сопровождении капитана Антонио, спасшего ему жизнь. Себастьян горюет о сестре, которая, по его мнению, погибла. Он хочет искать счастья при дворе герцога. Капитану больно расставаться с благородным юношей, к которому он успел искренне привязаться, но делать нечего — в Иллирии ему появляться опасно. Все же он тайно следует за Себастьяном, чтобы защитить его в случае нужды.
В доме Оливии сэр Тоби и сэр Эндрю в компании шута Фесте пьют вино и горланят песни. Мария пытается дружески урезонить их. Вслед за ней появляется дворецкий Оливии — чванливый зануда Мальволио. Он безуспешно старается прекратить пирушку. Когда дворецкий уходит, Мария всячески высмеивает этого «надутого осла», который «лопается от самодовольства», и клянется его одурачить. Она собирается написать ему любовное послание от имени Оливии и выставить на всеобщее посмешище.
Во дворце герцога шут Фесте сначала поет ему печальную песню о неразделенной любви, а потом пытается развеселить шутками. Орсино упивается своей любовью к Оливии, предыдущие неудачи его не обескураживают. Он убеждает Виолу опять отправиться к графине. Герцог высмеивает утверждение мнимого юноши, что какая-то женщина может быть влюблена в него так же сильно, как он в Оливию: «Грудь женщины не вынесет биения / Такой могучей страсти, как моя». Он остается глух ко всем намекам влюбленной Виолы.
Сэра Тоби и его сообщников просто распирает то от смеха, то от злости, когда они подслушивают, как Мальволио рассуждает о возможности брака со своей госпожой, о том, как он приструнит сэра Тоби, став в доме хозяином. Однако самая потеха начинается, когда дворецкий находит письмо, написанное Марией, подделавшей почерк Оливии. Мальволио быстро убеждает себя, что он-то и есть тот «безыменный возлюбленный», которому оно адресовано. Он решает неукоснительно следовать инструкциям, данным в письме и придуманным Марией специально в расчете на то, чтобы враг веселой компании вел себя и выглядел самым дурацким образом. Сэр Тоби в восторге от выдумки Марии, да и от нее самой: «За таким остроумнейшим дьяволенком хоть в самый Тартар».
В саду Оливии Виола и Фесте обмениваются остротами. «Он хорошо играет дурака. / Такую роль глупец не одолеет», — говорит Виола о шуте. Затем Виола говорит с вышедшей в сад Оливией, которая уже не скрывает своего страстного увлечения «юношей». Сэр Эндрю оскорблен тем, что в его присутствии графиня любезничала с герцогским слугой, и сэр Тоби убеждает его вызвать нахального юнца на поединок. Правда, сэр Тоби уверен, что у обоих не хватит мужества сразиться.
Антонио на городской улице встречается с Себастьяном и объясняет ему, что не может открыто сопровождать его, так как он участвовал в морском бою с галерами герцога и одержал верх — «меня узнают / И, уж поверьте, спуску не дадут». Себастьян хочет побродить по городу. Он уславливается с капитаном о встрече через час в лучшей гостинице. На прощание Антонио уговаривает друга принять его кошелек на случай неожиданных расходов.
Мальволио, глупо улыбающийся и безвкусно одетый (все по плану Марии), игриво цитирует Оливии пассажи из якобы ее послания. Оливия убеждена, что дворецкий спятил. Она поручает сэру Тоби позаботиться о нем, что тот и делает, только на свой лад: он сначала насмехается над несчастным спесивцем, а потом запихивает его в чулан. Затем принимается за сэра Эндрю и «Цезарио». Каждому он потихоньку говорит, что его противник свиреп и искусен в фехтовании, но избежать поединка невозможно. Наконец бледные от страха «дуэлянты» обнажают шпаги — и тут вмешивается проходящий мимо Антонио. Он закрывает собой Виолу, приняв ее за Себастьяна, и начинает драться с сэром Тоби, разъяренным тем, что его проделка не удалась. Появляются приставы. Они арестовывают Антонио по приказу герцога. Тот вынужден подчиниться, но просит у Виолы вернуть кошелек — деньги ему теперь понадобятся. Он возмущен тем, что человек, для которого он столько сделал, не узнает его и не хочет говорить ни о каких деньгах, хотя и благодарит за заступничество. Капитана уводят. Виола, понявшая, что ее спутали с Себастьяном, радуется спасению брата.
На улице сэр Эндрю набрасывается на своего противника, в робости которого недавно убедился, и дает ему пощечину, но... это не кроткая Виола, а отважный Себастьян. Трусоватый рыцарь крепко побит. Сэр Тоби пытается вступиться за него — Себастьян обнажает шпагу. Появившаяся Оливия останавливает драку и гонит дядю. «Цезарио, прошу вас, не сердитесь», — говорит она Себастьяну. Она ведет его в дом и предлагает обручиться. Себастьян растерян, но соглашается, красавица сразу очаровала его. Он хотел бы посоветоваться с Антонио, но тот куда-то пропал, в гостинице его нет. Тем временем шут, притворившись священником, долго разыгрывает сидящего в темном чулане Мальволио. Наконец, сжалившись, соглашается принести ему свечу и письменные принадлежности.
Перед домом Оливии герцог и Виола ожидают беседы с графиней. В это время приставы приводят Антонио, которого Виола называет «спасителем», а Орсино — «прославленным пиратом». Антонио горько укоряет Виолу в неблагодарности, хитрости и лицемерии. Из дома появляется Оливия. Она отвергает герцога, а «Цезарио» упрекает в неверности. Священник подтверждает, что два часа назад обвенчал графиню с герцогским любимцем. Орсино потрясен. Напрасно Виола говорит, что он стал ей «жизнью, светом», что он ей «милей всех женщин в мире этом», бедняжке никто не верит. Тут из сада появляются избитые сэр Тоби и сэр Эндрю с жалобами на герцогского придворного Цезарио, за ними с извинениями — Себастьян (незадачливая парочка опять нарвалась на мужчину). Себастьян видит Антонио и бросается к нему. И капитан, и герцог потрясены сходством близнецов. Они в полном недоумении. Брат и сестра узнают друг друга. Орсино, поняв, что тот, кто был ему так дорог в образе юноши, на самом деле влюбленная в него девушка, полностью примиряется с потерей Оливии, которую готов теперь считать сестрою. Ему не терпится увидеть Виолу в женском наряде: «...передо мной предстанет дева, — / Моей души любовь и королева». Шут приносит письмо Мальволио. Странности дворецкого получают объяснение, но Марию не наказывают за злую шутку — она теперь леди, сэр Тоби в благодарность за ее проделки женился на ней. Оскорбленный Мальволио покидает дом — единственный мрачный персонаж уходит со сцены. Герцог приказывает «догнать его и к мировой склонить». Пьеса заканчивается шутливо-меланхолической песенкой, которую поет Фесте.
