Действие происходит в Афинах. Правитель Афин носит имя Тесея, одного из популярнейших героев античных преданий о покорении греками воинственного племени женщин — амазонок. На царице этого племени, Ипполите, и женится Тесей. Пьеса, видимо, была создана для спектакля по случаю свадьбы каких-то высокопоставленных лиц.
Идут приготовления к свадьбе герцога Тесея и царицы амазонок Ипполиты, которая должна состояться в ночь полнолуния. Ко дворцу герцога является разгневанный Эгей, отец Гермии, который обвиняет Лизандра в том, что он околдовал его дочь и коварно заставил ее полюбить его, в то время как она уже обещана Деметрию. Гермия признается в любви к Лизандру. Герцог объявляет, что по афинским законам она должна подчиниться воле отца. Он дает девушке отсрочку, но в день новолуния ей придется «или умереть / За нарушение отцовской воли, / Иль обвенчаться с тем, кого он выбрал, / Иль дать навек у алтаря Дианы / Обет безбрачья и суровой жизни». Влюбленные договариваются вместе бежать из Афин и встретиться следующей ночью в ближайшем лесу. Они открывают свой план подруге Гермии Елене, которая когда-то была возлюбленной Деметрия и до сих пор любит его страстно. Надеясь на его благодарность, она собирается рассказать Деметрию о планах влюбленных. Тем временем компания простоватых мастеровых готовится к постановке интермедии по случаю свадьбы герцога. Режиссер, плотник Питер Пигва, выбрал подходящее произведение: «Прежалостная комедия и весьма жестокая кончина Пирама и Фисбы». Ткач Ник Основа согласен сыграть роль Пирама, как, впрочем, и большинство других ролей. Починщику раздувальных мехов Френсису Дудке дается роль Фисбы (во времена Шекспира женщины на сцену не допускались). Портной Робин Заморыш будет матерью Фисбы, а медник Том Рыло — отцом Пирама. Роль Льва поручают столяру Миляге: у него «память туга на учение», а для этой роли нужно только рычать. Пигва просит всех вызубрить роли наизусть и завтра вечером прийти в лес к герцогскому дубу на репетицию.
В лесу поблизости от Афин царь фей и эльфов Оберон и его жена царица Титания ссорятся из-за ребенка, которого Титания усыновила, а Оберон хочет забрать себе, чтобы сделать пажом. Титания отказывается подчиниться воле мужа и уходит вместе с эльфами. Оберон просит озорного эльфа Пэка (Доброго Малого Робина) принести ему маленький цветок, на который упала стрела Купидона, после того как он промахнулся «в царящую на Западе Весталку» (намек на королеву Елизавету). Если веки спящего смазать соком этого цветка, то, проснувшись, он влюбится в первое живое существо, которое увидит. Когда Пэк возвращается с цветком, Оберон поручает ему разыскать Деметрия, которого описывает как «надменного повесу» в афинских одеждах, и смазать ему глаза, но так, чтобы во время пробуждения с ним рядом оказалась влюбленная в него красавица. Обнаружив спящую Титанию, Оберон выжимает сок цветка на ее веки. Пэк, приняв Лизандра за Деметрия, капает сок ему на глаза. Появляется Елена, от которой убежал Деметрий, и остановившись отдохнуть, будит Лизандра, который тут же в нее влюбляется. Елена считает, что он насмехается над ней, и убегает, а Лизандр, бросив Гермию, устремляется за Еленой.
Рядом с местом, где спит Титания, собралась на репетицию компания мастеровых. По предложению Основы, который очень озабочен тем, чтобы, упаси Бог, не напугать дам-зрительниц, к пьесе пишут два пролога — первый о том, что Пирам вовсе не убивает себя и никакой он на самом деле не Пирам, а ткач Основа, а второй — что и Лев совсем не лев, а столяр Миляга. Шалун Пэк, который с интересом наблюдает за репетицией, заколдовывает Основу: теперь у ткача ослиная голова. Дружки, приняв Основу за оборотня, в страхе разбегаются. В это время просыпается Титания и, взглянув на Основу, говорит: «Твой образ пленяет взор <...> Тебя люблю я. Следуй же за мной!» Титания призывает четырех эльфов — Горчичное Зерно, Душистый Горошек, Паутинку и Мотылька — и приказывает им служить «своему милому». Оберон в восторге выслушивает рассказ Пэка о том, как Титания влюбилась в чудовище, но весьма недоволен, узнав, что эльф брызнул волшебным соком в глаза Лизандра, а не Деметрия. Оберон усыпляет Деметрия и исправляет ошибку Пэка, который по приказу своего властелина заманивает Елену поближе к спящему Деметрию. Едва проснувшись, Деметрий начинает клясться в любви той, которую Недавно с презрением отвергал. Елена же убеждена, что оба молодых человека, Лизандр и Деметрий, над ней издеваются: «Пустых насмешек слушать нету силы!» К тому же она считает, что Гермия с ними заодно, и горько корит подругу за коварство. Потрясенная грубыми оскорблениями Лизандра, Гермия обвиняет Елену в том, что она обмаящица и воровка, укравшая у нее сердце Лизандра. Слово за слово — и она уже пытается выцарапать Елене глаза. Молодые люди — теперь соперники, добивающиеся любви Елены, — удаляются, чтобы в поединке решить, кто из них имеет больше прав. Пэк в восторге от всей этой путаницы, но Оберон приказывает ему завести обоих дуэлянтов поглубже в лес, подражая их голосам, и сбить их с пути, «чтоб им никак друг друга не найти». Когда Лизандр в изнеможении сваливается с ног и засыпает, Пэк выжимает на его веки сок растения — противоядия любовному цветку. Елена и Деметрий также усыплены неподалеку друг от друга.
Увидев Титанию, уснувшую рядом с Основой, Оберон, который к этому времени уже заполучил понравившегося ему ребенка, жалеет ее и дотрагивается до ее глаз цветком-противоядием. Царица фей просыпается со словами: «Мой Оберон! Что может нам присниться! / Мне снилось, что влюбилась я в осла!» Пэк по приказу Оберона возвращает Основе его собственную голову. Повелители эльфов улетают. В лесу появляются охотящиеся Тесей, Ипполита и Эгей, Они находят спящих молодых людей и будят их. Уже свободный от действия любовного зелья, но все еще ошеломленный Лизандр объясняет, что они с Гермией бежали в лес от суровости афинских законов, Деметрий же признается, что «Страсть, цель и радость глаз теперь / Не Гермия, а милая Елена». Тесей объявляет, что еще две пары будут сегодня венчаться вместе с ними и Ипполитой, после чего удаляется вместе со свитой. Проснувшийся Основа отправляется в дом Пигвы, где его с нетерпением ждут друзья. Он дает актерам последние наставления: «Фисба пусть наденет чистое белье», а Лев пусть не вздумает обрезать ногти — они должны выглядывать из-под шкуры, как когти.
Тезей дивится странному рассказу влюбленных. «Безумные, любовники, поэты — / Все из фантазий созданы одних», — говорит он. Распорядитель увеселений Филострат представляет ему список развлечений. Герцог выбирает пьесу мастеровых: «Не может никогда быть слишком плохо, / Что преданность смиренно предлагает». Под иронические комментарии зрителей Пигва читает пролог. Рыло объясняет, что он — Стена, через которую переговариваются Пирам и Фисба, и потому измазан известкой. Когда Основа-Пирам ищет щель в Стене, чтобы взглянуть на возлюбленную, Рыло услужливо растопыривает пальцы. Появляется Лев и в стихах объясняет, что он не настоящий. «Какое кроткое животное, — восхищается Тесей, — и какое рассудительное!» Самодеятельные актеры безбожно перевирают текст и говорят массу глупостей, чем изрядно потешают своих знатных зрителей. Наконец пьеса закончена. Все расходятся — уже полночь, волшебный час для влюбленных. Появляется Пэк, он и остальные эльфы сначала поют и танцуют, а потом по распоряжению Оберона и Титании разлетаются по дворцу, чтобы благословить постели новобрачных. Пэк обращается к зрителям: «Коль я не смог вас позабавить, / Легко вам будет все исправить: / Представьте, будто вы заснули / И перед вами сны мелькнули».
